AMPK

AMPK (AMP-activated protein kinase) is an enzyme that works as the cell’s fuel gauge: it switches on when cellular energy runs low and shifts the cell from building things to burning fuel. It sits in essentially every cell of the body and in every eukaryote studied, from yeast to humans. Exercise, food restriction and drugs such as metformin all raise its activity, which is why the name turns up so often in longevity writing. What it measures

AMPK is a signaling protein, not a value on a lab report. It is assembled from three subunits: a catalytic alpha subunit and two regulatory ones, beta and gamma. The gamma subunit binds adenine nucleotides, and that is where the sensing happens. When ATP is plentiful the enzyme stays quiet; when ATP has been spent and its breakdown products AMP and ADP accumulate, they displace it, and the enzyme is switched on through phosphorylation of a site on the alpha subunit by an upstream kinase called LKB1. A second route, through the calcium-sensing kinase CaMKK2, activates it even when energy is not low.

Once active, AMPK phosphorylates a long list of targets, and the pattern is consistent: it turns on processes that generate ATP — glucose uptake into muscle, fatty acid oxidation, the building of new mitochondria — and turns off processes that consume it, such as the synthesis of fatty acids, cholesterol and protein. Two of those targets connect it to the rest of the longevity vocabulary. It restrains mTOR, the opposing sensor that drives growth when nutrients are abundant, and it directly activates ULK1, the kinase that starts Autophagy.

There is no clinical test. In research it is read from tissue, usually a muscle biopsy, by detecting the phosphorylated active form of the protein. No blood panel, wearable or supplement label tells you what your AMPK is doing.

Why it matters for longevity

In short-lived laboratory species the evidence is genetic and reasonably strong. Raising AMPK activity extends lifespan in the roundworm C. elegans and in fruit flies, and a working copy of the enzyme is required for some interventions that extend life in those animals, including dietary restriction. Remove it and the intervention stops working.

In mammals the picture is less settled. AMPK activity appears to fall with age in several tissues, and low activity travels with obesity, inactivity and insulin resistance, but no study has shown that raising it lengthens life in a mammal. Human evidence is entirely indirect: the things that activate AMPK — regular training, sustained weight loss, metformin in people with diabetes — also carry good outcome data, and that association does not establish AMPK as the reason. More activation is also not automatically better. The same energy-sparing response helps stressed tumor cells survive, and because AMPK restrains mTOR, chronically pushing it up would work against the muscle growth that matters in later life.

What changes it

Exercise is the most reliable activator, and the effect scales with how hard and how long you work and with how depleted your muscle glycogen is. Any sustained energy deficit does the same: caloric restriction, weight loss and long gaps between meals lower the cell’s energy charge and lift AMPK. Sleep loss and chronic overfeeding push in the opposite direction.

On the drug side, metformin and the plant alkaloid berberine both raise AMPK activity indirectly, by mildly inhibiting complex I of the mitochondrial respiratory chain so that AMP and ADP build up; how much of their clinical effect actually runs through AMPK is still argued. Salicylate activates it directly, and direct activators are in pharmaceutical development. Supplements marketed as AMPK activators are worth reading skeptically: the concentrations used in cell culture are often far above what oral dosing achieves in a person, and nobody can measure whether it worked in you.
