What cellular senescence is

Senescent cells are cells that have permanently stopped dividing but refuse to die, and they leak inflammatory, tissue-damaging signals into the space around them — which is why they are nicknamed “zombie cells”. Senolytics are compounds designed to kill those cells selectively while leaving healthy cells alone; in mice they delayed age-related disease, and in humans only the first small trials have been run so far. When a cell is badly damaged, or has divided many times, it can enter a state of Cellular senescence: it shuts down division but stays alive. Early in life this is protective, including against cancer, and the immune system usually clears such cells. With age that clearance works less well, and senescent cells accumulate. Why they cause harm

The problem is not that these cells simply sit there doing nothing. They secrete a cocktail of pro-inflammatory and tissue-degrading molecules known as the senescence-associated secretory phenotype (SASP). Through the SASP, even a small number of senescent cells sustains chronic low-grade inflammation (see our article on inflammaging) and pushes neighboring cells toward the same state. Hence the “zombie cell” label. Senolytics: the idea and the evidence

Senolytics are compounds that force senescent cells to die — to undergo Apoptosis — without touching healthy ones. The logic is straightforward: remove the source of the harmful signals. Here is what is known:

  • in experiments, clearing senescent cells in mice delayed age-related disease and increased Healthspan (healthy life expectancy);
  • the combination of dasatinib and quercetin, and a preparation based on the flavonoid fisetin, improved tissue function in animal experiments;
  • in humans, the first phases of trials are under way (in pulmonary fibrosis and aging-related conditions, for example), and at this stage they test safety and feasibility rather than benefit.

In aging biology, impressive results in mice often fail to replicate in humans. Senolytics are a promising direction, but not a proven one.

Why you should not experiment on yourself

“Senolytic protocols” are sold online, but buying one is premature and risky. Dasatinib is a serious anticancer drug with real side effects, and doses, schedules and safety in healthy people have not been established. Supplements containing fisetin or quercetin are marketed as senolytics, yet there is no evidence of clinical benefit in humans. The sensible position is to follow the trial results rather than run ahead of the science.
